Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.2255A>G (p.Tyr752Cys), citing Ambry Variant Classification Scheme 2023: The p.Y752C variant (also known as c.2255A>G), located in coding exon 28 of the CACNA2D1 gene, results from an A to G substitution at nucleotide position 2255. The tyrosine at codon 752 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
