Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.647G>A (p.Arg216Gln), citing Ambry Variant Classification Scheme 2023: The p.R216Q variant (also known as c.647G>A), located in coding exon 7 of the CACNA2D1 gene, results from a G to A substitution at nucleotide position 647. The arginine at codon 216 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
