NM_005751.5(AKAP9):c.9482_9484dup (p.Val3161_Ala3162insVal) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.9482_9484dupTTG variant (also known as p.V3161dup), located in coding exon 39 of the AKAP9 gene, results from an in-frame duplication of TTG at nucleotide positions 9482 to 9484. This results in the duplication of an extra residue between codons 3161 and 3162. This amino acid position is not well conserved in available vertebrate species. In addition, this variant is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
