Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.1517C>T (p.Thr506Ile), citing Ambry Variant Classification Scheme 2023: The p.T506I variant (also known as c.1517C>T), located in coding exon 13 of the CHEK2 gene, results from a C to T substitution at nucleotide position 1517. The threonine at codon 506 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
