Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.9724A>G (p.Lys3242Glu), citing Ambry Variant Classification Scheme 2023: The p.K3242E variant (also known as c.9724A>G), located in coding exon 59 of the DNAH11 gene, results from an A to G substitution at nucleotide position 9724. The lysine at codon 3242 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
