Pathogenic — the classification assigned by Ambry Genetics to NM_006885.4(ZFHX3):c.2095_2096delinsTGC (p.Gly699fs), citing Ambry Variant Classification Scheme 2023: The c.2095_2096delGGinsTGC (p.G699Cfs*34) alteration, located in exon 2 (coding exon 1) of the ZFHX3 gene, consists of an deletion of 2 and insertion of 3 nucleotides causing a translational frameshift at position 2095 with a predicted alternate stop codon after 34 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
