Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_031443.4(CCM2):c.431A>G (p.Tyr144Cys), citing Ambry Variant Classification Scheme 2023: The p.Y144C variant (also known as c.431A>G), located in coding exon 4 of the CCM2 gene, results from an A to G substitution at nucleotide position 431. The tyrosine at codon 144 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
