This variant lies in the BARD1 gene (transcript NM_000465.4) at coding-DNA position 1810, where G is replaced by A; at the protein level this means replaces valine at residue 604 with isoleucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This sequence change replaces valine with isoleucine at codon 604 of the BARD1 protein (p.Val604Ile). The valine residue is highly conserved and there is a small physicochemical difference between valine and isoleucine. This variant also falls at the last nucleotide of exon 8, which is part of the consensus splice site for this exon. This variant is not present in population databases (ExAC no frequency).
