Pathogenic for Juvenile polyposis syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000018.9:g.(?_48573411)_(48604843_?)del, citing Invitae Variant Classification Sherloc (09022015): A gross deletion of the genomic region encompassing the full coding sequence of the SMAD4 gene has been identified. The boundaries of this event are unknown as the deletion extends beyond the assayed region for this gene and therefore may encompass additional genes. Loss-of-function variants in SMAD4 are known to be pathogenic. A deletion of the entire SMAD4 gene has been reported in the literature in individuals affected with juvenile polyposis syndrome (PMID: 17873119). For these reasons, this variant has been classified as Pathogenic.
