Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001211.6(BUB1B):c.1189A>G (p.Met397Val), citing Ambry Variant Classification Scheme 2023: The p.M397V variant (also known as c.1189A>G), located in coding exon 9 of the BUB1B gene, results from an A to G substitution at nucleotide position 1189. The methionine at codon 397 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
