Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251476 control chromosomes. To our knowledge, no occurrence of c.280C>T in individuals affected with Glutaric Acidemia Type 1 and no experimental evidence demonstrating its impact on protein function have been reported. Different variants affecting the same codon have been classified as pathogenic by our lab (c.281G>A, p.Arg94Gln & c.281G>T, p.Arg94Leu), supporting the critical relevance of codon 94 to GCDH protein function. ClinVar contains an entry for this variant (Variation ID: 459950). Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
