Uncertain significance for Ehlers-Danlos syndrome, kyphoscoliotic type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000302.4(PLOD1):c.1145A>C (p.Asp382Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid with alanine at codon 382 of the PLOD1 protein (p.Asp382Ala). The aspartic acid residue is highly conserved and there is a moderate physicochemical difference between aspartic acid and alanine. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a PLOD1-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance.
