Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.752A>G (p.Lys251Arg), citing Ambry Variant Classification Scheme 2023: The p.K251R variant (also known as c.752A>G), located in coding exon 10 of the COL3A1 gene, results from an A to G substitution at nucleotide position 752. The lysine at codon 251 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved through mammals, but arginine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
