Uncertain significance — the classification assigned by Ambry Genetics to NM_001127208.3(TET2):c.1076A>T (p.Asn359Ile), citing Ambry Variant Classification Scheme 2023: The p.N359I variant (also known as c.1076A>T), located in coding exon 1 of the TET2 gene, results from an A to T substitution at nucleotide position 1076. The asparagine at codon 359 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
