Uncertain significance for Loeys-Dietz syndrome 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003238.6(TGFB2):c.334T>C (p.Phe112Leu), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with leucine at codon 112 of the TGFB2 protein (p.Phe112Leu). The phenylalanine residue is moderately conserved and there is a small physicochemical difference between phenylalanine and leucine. This variant has not been reported in the literature in individuals with a TGFB2-related disease. In summary, this variant has uncertain impact on TGFB2 function. The available evidence is currently insufficient to determine its role in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The leucine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
