This variant lies in the EYA1 gene (transcript NM_000503.6) at coding-DNA position 1684, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 562 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: Family studies indicate this nonsense variant likely was not inherited from either parent (i.e. occurred de novo) in an individual with disease (Invitae database). For these reasons, this variant has been classified as Pathogenic. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the penultimate exon of the EYA1 mRNA at codon 562 (p.Gln562*). While this is not anticipated to result in nonsense-mediated decay, it is expected to create a truncated EYA1 protein.
