Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.2943G>T (p.Glu981Asp), citing Ambry Variant Classification Scheme 2023: The p.E981D variant (also known as c.2943G>T), located in coding exon 23 of the FBN2 gene, results from a G to T substitution at nucleotide position 2943. The glutamic acid at codon 981 is replaced by aspartic acid, an amino acid with highly similar properties, and is located in the TGFBP #03 domain. This amino acid position is well conserved in available vertebrate species; however, aspartic acid is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
