Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_015338.6(ASXL1):c.997A>C (p.Met333Leu), citing Ambry Variant Classification Scheme 2023: The p.M333L variant (also known as c.997A>C), located in coding exon 11 of the ASXL1 gene, results from an A to C substitution at nucleotide position 997. The methionine at codon 333 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
