NM_000546.6(TP53):c.156dup (p.Trp53fs) was classified as Pathogenic for Li-Fraumeni syndrome by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, loss-of-function variants in TP53 are known to be pathogenic (PMID: 20522432). This sequence change inserts 1 nucleotide in exon 4 of the TP53 mRNA (c.156dupA), causing a frameshift at codon 53. This creates a premature translational stop signal (p.Trp53Metfs*4) and is expected to result in an absent or disrupted protein product.
