Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000546.6(TP53):c.1165G>A (p.Gly389Arg), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TP53 c.1165G>A (p.Gly389Arg) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant allele was found at a frequency of 3.2e-05 in 31400 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1165G>A in individuals affected with TP53-related conditions has been reported. The IARC TP53 database reports this variant to be functional in transcriptional activation activity based on 8 different promoters as measured by yeast assays (Kato_2003). The following publication have been ascertained in the context of this evaluation (PMID: 12826609). ClinVar contains an entry for this variant (Variation ID: 458522). Based on the evidence outlined above, the variant was classified as VUS-possibly benign.
