Uncertain significance for Werner syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000553.6(WRN):c.2081T>C (p.Leu694Pro), citing Invitae Variant Classification Sherloc (09022015): This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a WRN-related disease. In summary, this variant is a novel missense change with uncertain impact on protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies. This sequence change replaces leucine with proline at codon 694 of the WRN protein (p.Leu694Pro). The leucine residue is highly conserved and there is a moderate physicochemical difference between leucine and proline.
