This variant lies in the MPL gene (transcript NM_005373.3) at coding-DNA position 212, where G is replaced by A; at the protein level this means replaces arginine at residue 71 with glutamine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 71 of the MPL protein (p.Arg71Gln). This variant also falls at the last nucleotide of exon 2, which is part of the consensus splice site for this exon. This variant is present in population databases (rs368753117, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with MPL-related conditions. ClinVar contains an entry for this variant (Variation ID: 458371). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
