NM_001114753.3(ENG):c.904dup (p.Glu302fs) was classified as Pathogenic for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.904dupG pathogenic mutation, located in coding exon 7 of the ENG gene, results from a duplication of G at nucleotide position 904, causing a translational frameshift with a predicted alternate stop codon (p.E302Gfs*32). This mutation was seen in a patient with telangiectasia, pulmonary AVMs, and two episodes of stroke (Cymerman U et al. Hum. Mutat., 2003 May;21:482-92). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
