Pathogenic for Hereditary hemorrhagic telangiectasia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000009.12:g.(?_127829681)_(127829833_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exon(s) 3 of the ENG gene. This variant would be expected to be in-frame, preserving the integrity of the reading frame. A similar copy number variant has been observed in individuals with hemorrhagic telangiectasia (PMID: 20412114, 20414677). Algorithms developed to predict the effect of variants on protein structure and function are not available or were not evaluated for this variant. Experimental studies have shown that a similar copy number variant affects ENG protein function (PMID: 9366572). For these reasons, this variant has been classified as Pathogenic.
