This variant lies in the CTSD gene (transcript NM_001909.5) at coding-DNA position 271, where T is replaced by C; at the protein level this means replaces cysteine at residue 91 with arginine — a missense variant. Submitter rationale: This sequence change replaces cysteine with arginine at codon 91 of the CTSD protein (p.Cys91Arg). The cysteine residue is moderately conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency).
