Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014915.3(ANKRD26):c.3769C>A (p.Gln1257Lys), citing Ambry Variant Classification Scheme 2023: The p.Q1257K variant (also known as c.3769C>A), located in coding exon 25 of the ANKRD26 gene, results from a C to A substitution at nucleotide position 3769. The glutamine at codon 1257 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
