NM_000268.4(NF2):c.932G>A (p.Arg311Lys) was classified as Uncertain Significance for Neurofibromatosis, type 2 by All of Us Research Program, National Institutes of Health, citing ACMG Guidelines, 2015. This variant lies in the NF2 gene (transcript NM_000268.4) at coding-DNA position 932, where G is replaced by A; at the protein level this means replaces arginine at residue 311 with lysine — a missense variant. Submitter rationale: This missense variant replaces arginine with lysine at codon 311 of the NF2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with NF2-related disorders in the literature. This variant has been identified in 1/251478 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
