Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042492.3(NF1):c.8066del (p.Val2689fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 8066, deleting one base; at the protein level this means shifts the reading frame starting at valine residue 2689, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change deletes 1 nucleotide from exon 54 of the NF1 mRNA (c.8003delT), causing a frameshift at codon 2668. This creates a premature translational stop signal (p.Val2668Glyfs*50) and is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538).
