Pathogenic for Neurofibromatosis, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001042492.3(NF1):c.5013_5017dup (p.Asn1673fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change duplicates 5 nucleotides in exon 36 of the NF1 mRNA (c.4950_4954dupCGACA), causing a frameshift at codon 1652. This creates a premature translational stop signal (p.Asn1652Thrfs*27) and is expected to result in an absent or disrupted protein product. While this particular variant has not been reported in the literature, loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). For these reasons, this variant has been classified as Pathogenic.
