This variant lies in the NF1 gene (transcript NM_001042492.3) at coding-DNA position 3374, where C is replaced by T; at the protein level this means replaces alanine at residue 1125 with valine — a missense variant. Submitter rationale: The NF1 c.3374C>T (p.A1125V) variant has been reported in heterozygosity in at least one individual being evaluated for neurofibromatosis, type 1 (PMID: 10336779). In breast cancer case-control studies, it has only been observed in the control populations (PMID: 30287823, 33471991). It was observed in 2/30616 chromosomes of the South Asian subpopulation in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID 457644). In silico tools suggest the impact of the variant on protein function is benign, though these predictions have not been confirmed by functional studies. There is no indication that this variant causes disease, but the evidence is insufficient currently to prove that conclusively. Thus, the clinical significance of this variant is currently uncertain.
