Uncertain significance — the classification assigned by Ambry Genetics to NM_006506.5(RASA2):c.983C>T (p.Pro328Leu), citing Ambry Variant Classification Scheme 2023: The p.P328L variant (also known as c.983C>T), located in coding exon 10 of the RASA2 gene, results from a C to T substitution at nucleotide position 983. The proline at codon 328 is replaced by leucine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
