This variant lies in the RAD50 gene (transcript NM_005732.4) at the canonical splice donor site of the intron immediately after coding-DNA position 213, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 2 of the RAD50 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in a shortened protein product. This variant is present in population databases (rs765484171, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with RAD50-related conditions.
