NM_001370259.2(MEN1):c.244G>C (p.Asp82His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D82H variant (also known as c.244G>C), located in coding exon 1 of the MEN1 gene, results from a G to C substitution at nucleotide position 244. The aspartic acid at codon 82 is replaced by histidine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
