NM_000138.5(FBN1):c.3295G>A (p.Glu1099Lys) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1099K variant (also known as c.3295G>A), located in coding exon 26 of the FBN1 gene, results from a G to A substitution at nucleotide position 3295. The glutamic acid at codon 1099 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
