NM_005751.5(AKAP9):c.6343G>T (p.Ala2115Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A2115S variant (also known as c.6343G>T), located in coding exon 27 of the AKAP9 gene, results from a G to T substitution at nucleotide position 6343. The alanine at codon 2115 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
