NM_000719.7(CACNA1C):c.3235G>A (p.Gly1079Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1079R variant (also known as c.3235G>A), located in coding exon 26 of the CACNA1C gene, results from a G to A substitution at nucleotide position 3235. The glycine at codon 1079 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
