While this particular variant has not been reported in the literature, loss-of-function variants in COL1A1 are known to be pathogenic (PMID: 9443882, 9295084, 7942841). This sequence change deletes 1 nucleotide from exon 32 of the COL1A1 mRNA (c.2217delA), causing a frameshift at codon 741. This creates a premature translational stop signal (p.Pro741Leufs*25) and is expected to result in an absent or disrupted protein product.
