NM_001166108.2(PALLD):c.1965-12790T>G was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W81G variant (also known as c.241T>G), located in coding exon 1 of the PALLD gene, results from a T to G substitution at nucleotide position 241. The tryptophan at codon 81 is replaced by glycine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
