Submitter rationale: The c.706-3C>A intronic variant results from a C to A substitution 3 nucleotides upstream from coding exon 7 in the PMS2 gene. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration may weaken the native splice acceptor site; however, direct evidence is insufficient at this time (Ambry internal data).
