This variant lies in the PMS2 gene (transcript NM_000535.7) at coding-DNA position 253, where C is replaced by G; at the protein level this means replaces leucine at residue 85 with valine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces leucine with valine at codon 85 of the PMS2 protein (p.Leu85Val).
