Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.2235A>G (p.Ile745Met), citing Ambry Variant Classification Scheme 2023: The p.I745M variant (also known as c.2235A>G), located in coding exon 13 of the PMS2 gene, results from an A to G substitution at nucleotide position 2235. The isoleucine at codon 745 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
