Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4024C>G (p.Gln1342Glu), citing Ambry Variant Classification Scheme 2023: The p.Q1342E variant (also known as c.4024C>G), located in coding exon 33 of the TSC2 gene, results from a C to G substitution at nucleotide position 4024. The glutamine at codon 1342 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
