While this particular variant has not been reported in the literature, loss-of-function variants in MSH6 are known to be pathogenic (PMID: 24362816, 18269114). This sequence change deletes 8 nucleotides from exon 9 of the MSH6 mRNA (c.3854_3861delTCCTCTAT) and creates a premature translational stop signal at codon 1285 (p.Phe1285*) of the MSH6 gene. It is expected to result in an absent or disrupted protein product.
