This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3141, where G is replaced by C; at the protein level this means replaces tryptophan at residue 1047 with cysteine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 455234). This variant has not been reported in the literature in individuals affected with MSH6-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces tryptophan with cysteine at codon 1047 of the MSH6 protein (p.Trp1047Cys).
