Uncertain significance for Hereditary nonpolyposis colorectal neoplasms — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000179.3(MSH6):c.2582C>T (p.Ala861Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 2582, where C is replaced by T; at the protein level this means replaces alanine at residue 861 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 861 of the MSH6 protein (p.Ala861Val). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with Lynch syndrome-related cancers (PMID: 23729658). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 455207). Invitae Evidence Modeling incorporating data from in vitro experimental studies (internal data) indicates that this missense variant is not expected to disrupt MSH6 function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
