Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000455.5(STK11):c.46G>T (p.Glu16Ter), citing Ambry Variant Classification Scheme 2023: The p.E16* variant (also known as c.46G>T), located in coding exon 1 of the STK11 gene, results from a G to T substitution at nucleotide position 46. This changes the amino acid from a glutamic acid to a stop codon within coding exon 1. The predicted stop codon occurs in the 5&rsquo; end of thegene. Premature termination codons in the 5&rsquo; end of a gene have been reported to escape nonsense-mediated mRNAdecay and/or lead to re-initiation (Rivas et al.
