Pathogenic for Hereditary nonpolyposis colon cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000007.14:g.(?_5986753)_(5997429_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is an out-of-frame deletion of the genomic region encompassing exons 7-11 of the PMS2 gene. This is expected to create a premature translational stop signal and result in an absent or disrupted protein product. A similar deletion of exon 7-11 has been observed in individual(s) with constitutional mismatch repair deficiency (PMID: 23582141). In at least one individual the data is consistent with the variant being in trans (on the opposite chromosome) from a pathogenic variant. Loss-of-function variants in PMS2 are known to be pathogenic (PMID: 21376568, 24362816). For these reasons, this variant has been classified as Pathogenic.
