Pathogenic for Hereditary nonpolyposis colon cancer — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000003.11:g.(?_37034542)_(37050402_?)del, citing Invitae Variant Classification Sherloc (09022015): This variant is a gross deletion of the genomic region encompassing exons 1-6 of the MLH1 gene, which includes the initiator codon. The 5' end of this event is unknown as it extends beyond the assayed region for this gene and therefore may encompass additional genes. The 3' boundary is likely confined to intron 5 of the MLH1 gene. This is expected to result in an absent or disrupted protein product. Deletions of exons 1-6 have been reported in individuals affected with Lynch syndrome and colorectal cancer (PMID: 14635101, 19324997). Loss-of-function variants in MLH1 are known to be pathogenic (PMID: 15713769, 24362816). For these reasons, this variant has been classified as Pathogenic.
