NM_178452.6(DNAAF1):c.715del (p.Ser239fs) was classified as Pathogenic for Primary ciliary dyskinesia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, loss-of-function variants in DNAAF1 are known to be pathogenic (PMID: 19944405, 19944400). This sequence change deletes 1 nucleotide from exon 5 of the DNAAF1 mRNA (c.715delA), causing a frameshift at codon 239. This creates a premature translational stop signal (p.Ser239Alafs*12) and is expected to result in an absent or disrupted protein product.
