Submitter rationale: The p.E432K variant (also known as c.1294G>A), located in coding exon 8 of the DNAAF1 gene, results from a G to A substitution at nucleotide position 1294. The glutamic acid at codon 432 is replaced by lysine, an amino acid with similar properties. This variant was identified in one individual with with levocardia, recurrent respiratory illness, and ciliary dysfunction; no second DNAAF1 variant was reported (Nakhleh N et al. Circulation, 2012 May;125:2232-42). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
